ABSTRACT

To investigate effectsof Yangyinyiqi Mixture on pulmonary fibrosis caused by bleomycin. SD ratswere divided randomly into: model group(distilled water,1 mL·0.1 kg-1), dexamethasone acetate group (dexamethasone acetate, the dosage was reduced gradually), low-dose group (Yangyinyiqi Mixture, 11 g·kg-1), moderate-dose group (Yangyinyiqi Mixture, 22 g·kg-1), high-dose group (Yangyinyiqi Mixture, 44 g·kg-1) and control group (distilled water, 1 mL·0.1 kg-1). Yangyinyiqi Mixture and dexamethasone acetate were intragastrically administrated. Lung tissue was collected for histopathological examination. Compared with control group, collagen markedly increased and HYP content significantly increased on 7th day in model group (p<0.01). On 28th day, collagen was diffusely deposited, alveolar was destroyed, and HYP content significantly increased (p<0.01). Compared with model group, bleomycin-induced suffering injury caused MMP-9 expression levels to rapidly increase (7and 14 days, p<0.01). TIMP-1 markedly increased (7and 14 days, p<0.01) and stayed at a high level to28th day. Yangyinyiqi Mixture exerted an effect against pulmonary fibrosis, which could involved prevention of collagen deposition through inhibitingMMP-9 and TIMP-1 expression.


El trabajo investiga los efectos de la mezcla Yangyinyiqi sobre la fibrosis pulmonary causada por bleomicina. Ratas SD se dividieron aleatoriamente en: grupo modelo (agua destilada, 1 mL·0.1 kg-1), grupo acetate de dexametasona (acetate de dexametasona, la dosis se redujo gradualmente), grupo de dosis baja (mezcla Yangyinyiqi, 11 g·kg-1), grupo de dosis moderada (mezcla Yangyinyiqi, 22 g·kg-1), grupo de dosis alta (mezcla Yangyinyiqi, 44 g·kg-1) y grupo control (agua destilada, 1 Ml·0.1 kg-1). La mezcla de Yangyinyiqi y el acetate de dexametasona se administraron por vía intragástrica. Se recolectó tejido pulmonary para examen histopatológico. En comparación con el grupo control, el colágeno aumentó notablemente y el contenido de HYP aumentó significativamente el séptimo día en el grupo modelo (p<0.01). El día 28, el colágeno se depositó difusamente, se produjo destrucción alveolar y el contenido de HYP aumento significativamente (p<0.01). En comparación con el grupo modelo, la lesión inducida por bleomicina causó que los niveles de expression de MMP-9 aumentaron rápidamente (7 y 14 días, p<0.01). TIMP-1 aumentó notablemente (7 y 14 días, p<0.01) y se mantuvo en un nivel alto hasta el día 28. La mezcla Yangyinyiqi ejerció un efecto contra la fibrosis pulmonary, lo que podría implicar la prevención del deposito de colágenio mediante la inhibición de la expression de MMP-9 y TIMP-1.

ABSTRACT

Objective:To compare the effect of TiO 2 nanoparticles on antioxidant function and element content of liver and kidney tissues in young and adult rats .Methods:Forty-eight SD male rats , half in 4-week (youth) old and half in 9-week (adult) old rats, were randomly divided into 8 groups, which were exposed to TiO2 nanoparticles [(75 ±15) nm, anatase] through intragastric administration at 0, 10, 50 and 200 mg/kg body weight every day for 30 days.The liver and kidney tissues were collected for antioxidant function and element content analysis .Results: 200 mg/kg TiO2 nanoparticles exposure significantly increased the liver total superoxide dismutase ( T-SOD ) activity and the kidney reduced glutathione (GSH)/oxidized glutathione (GSSG) ratios in young rats, and significantly decreased the liver Mo, Co, Mn and P contents and the kidney Rb and Na contents in young rats .200 mg/kg TiO2 nanoparticles exposure significantly increased GSH/GSSG ratios and Rb contents and decreased Na con-tents in the liver of adult rats .No significantly difference was found in antioxidant indexes and elements content in the kidney of adult rats between three experimental groups and control group .Conclusion:TiO2 nanoparticles can enhance the antioxidant capacity and decrease the elements content in rat liver and kidney tissues .The liver is the more sensitive target organ and the young animals are more susceptible to TiO2 nanoparticles toxicity by the oral routes .

ABSTRACT

<p><b>OBJECTIVE</b>To explore the genotoxic effects of oral-exposed TiO2 nanoparticles on bone marrow cells in young rats.</p><p><b>METHODS</b>Twenty-eight SD male young rats (4 weeks old) were randomly divided into 4 groups, which were exposed to TiO2 nanoparticles ((75 ± 15) nm, anatase) through intragastric administration at 0, 10, 50 and 200 mg/kg body weight (bw) every day for 30 days. The bone marrow cells were collected for micronuclei and γ-H2AX immunofluorescence analysis.</p><p><b>RESULTS</b>The percentage of γ-H2AX foci-positive cells (37.4 ± 10.0)% in the 50 mg/kg bw dose group were significantly higher than that in the control group (19.8 ± 3.1)% (t value was -17.59, P < 0.01). No significantly difference was found in polychromatic erythrocyte/normochromatic erythrocyte (PCE/NCE) ratio and PCE micronucleus rate between three experimental groups and control group.</p><p><b>CONCLUSION</b>TiO2 nanoparticles can increase the frequency of DNA double-strand breaks in bone marrow cells, but has no effect on micronucleus of bone marrow cells in young rats.</p>

ABSTRACT

<p><b>OBJECTIVE</b>This study aimed to explore the impact of specimen collection and storage consumable products on trace element quantitative analysis.</p><p><b>METHODS</b>Devices and consumable products of different brands used in specimen collection or storage were selected and treated separately as below:urine collection and storage tubes (Brand A, B, C and D, 2 samples for each brand) were treated with 1% of HNO(3) volume fraction for 2 - 4 h; blood taking device (Brand O, P and Q, 3 samples for each brand) were used for ultra-pure water samples collecting as simulation of blood sampling;dust sampling filters (Brand X, Y and Z, 2 samples for each brand) were cold digested by nitric acid for 12 h, followed by microwave digestion. Then cadmium, cobalt, chromium, copper, iron, manganese, molybdenum, nickel, lead, selenium, stannum, titanium, vanadium and zinc concentrations in the solutions obtained during the course of collect or storage were quantified by inductively coupled plasma mass spectrometer.</p><p><b>RESULTS</b>For the urine collection and storage consumable products, background values of elements were described as mean of parellel samples. The consentration of 14 quantified elements were relatively low for 5 ml cryogenic vials (brand B) with background values range of 0.001 - 0.350 ng/ml. The background values of copper of 50 ml centrifuge tubes (brand A), chromium of 5 ml cryogenic vials (brand C) and zinc of 1.5 ml centrifuge tubes (brand D) were relatively high, which were 1.900, 1.095 and 1.368 ng/ml, respectively. Background values of elements in blood sampling devices were described as x(-) ± s. Background values of chromium for brand O, P and Q were (0.120 ± 0.017), (0.337 ± 0.093) and (0.360 ± 0.035) ng/ml; for copper were (0.050 ± 0.001), (0.017 ± 0.012) and (0.103 ± 0.015) ng/ml; for lead were (0.057 ± 0.072), (0.183 ± 0.118) and (0.347 ± 0.006) ng/ml; for titanium were (7.883 ± 0.145), (8.863 ± 0.190) and (8.613 ± 0.274) ng/ml; zinc were (2.240 ± 0.573), (42.140 ± 22.756) and (8.850 ± 3.670) ng/ml. There were statistically differences of background values for chromium, copper, lead, titanium and zinc among the above three brands of blood sampling devices (all P values < 0.05). For air sampling filters, background values of elements were described as mean of parellel samples. Background values of chromium and nickel of sampling filters (brand X) were lowest, which were 17.000 and 15.400 ng per piece, respectively; while background values for other elements were relatively high, the quantification of cadmium, cobalt, copper, iron, manganese, molybdenum, lead, selenium, stannum, titanium, vanadium and zinc were 0.250, 0.550, 48.500, 690.000, 25.500, 0.900, 6.500, 10.550, 7.950, 10.500, 0.850, 370.000 ng per piece, respectively. Background values of chromium and nickel of sampling filters (brand Z) were highest, which were 171.000 and 29.850 ng per piece.</p><p><b>CONCLUSION</b>Background values of trace elements varied among products of different brands, and the most noticable differences were found in chromium, manganese, nickel, lead, stannum and zinc.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the effect of titanium dioxide (TiO₂) nanoparticles on hemogram in rats with gastric ulcer.</p><p><b>METHODS</b>Physicochemical properties of TiO₂ nanoparticles were characterized. Twenty-four clear class SD male rats, aging 8 week-old, were randomly divided into 4 groups, 6 rats for each group. 20% acetic acid were injected into the rats' stomach on the border of gastric body and pyloric antrum, and hereby established the gastric ulcer model. The rats in 4 groups were exposed to TiO₂ nanoparticles through intragastric administration at 0, 10, 50 and 200 mg/kg body weight respectively for 30 days. Afterwards, the rats were conducted blood routine test and blood coagulation test for analysis.</p><p><b>RESULTS</b>TiO₂ nanoparticles were anatase crystals, closely spherical shape, whose average grain diameter was (75 ± 15) nm. The levels of white blood cell (WBC) count ((8.48 ± 3.28)×10⁹/L), lymphocyte (LYM) ((6.85 ± 2.53)×10⁹/L), monocyte (MOD) ((0.27 ± 0.12)×10⁹/L), granulocyte (GRN) ((1.37 ± 0.86)×10⁹/L), red blood cell (RBC) ((8.20 ± 0.49)×10⁹/L) and hematocrit (HCT) ((45.3 ± 1.4)%) in the 200 mg/kg dose group were significantly higher than those in the control group ((2.63 ± 0.34)×10⁹/L, (2.25 ± 0.26)×10⁹/L, (0.05 ± 0.06)×10⁹/L, (0.33 ± 0.26)× 10⁹/L, (4.87 ± 2.37)×10⁹/L and (27.2 ± 13.3)%, respectively; t values were -3.449, -3.825, -3.554, -3.097, -2.972 and -2.936 respectively, P values all < 0.05). The levels of WBC ((6.88 ± 3.06)×10⁹/L), MOD ((0.20 ± 0.07)×10⁹/L), RBC ((7.79 ± 0.48)×10⁹/L) and HCT ((42.7 ± 2.8)%) in 50 mg/kg dose group were also statistically higher than those in the control group (t values were -2.507, -2.367, -2.605 and -2.511 respectively, all P values < 0.05). There was no statistically difference found in other blood routine index and coagulation index between the three experimental groups and control group.</p><p><b>CONCLUSION</b>The long term intake of TiO₂ nanoparticles caused a statistically increase in the amount of WBC and RBC in rats with gastric ulcer; however, there was no obvious changes found in blood platelet and coagulation index.</p>

ABSTRACT

OBJECTIVE@#To analyze the significance of forensic autopsy in medical tangle.@*METHODS@#Ninety autopsy cases of medical legal dispute were retrospectively analyzed from the database of our department from 2001 to 2008. All cases were analyzed and classified based on age, sex, cause of death, clinic diagnosis and forensic diagnosis.@*RESULTS@#The age ranged from 1 day to 72 years, and the ratios of male to female is 1:1. The most common healthcare facilities involved were county hospitals (30 cases, 33.33%). The coincidence rate between clinical diagnoses and pathological diagnoses was 33.33%.@*CONCLUSION@#The forensic autopsy is valuable to solve or even avoid the occurrence of medical legal dispute.

ABSTRACT

OBJECTIVE@#To explore the effects of curcumin on the content of malondialdehyde (MDA) and the expression level of c-fos protein following hypoxia ischemia brain damage (HIBD) in rats.@*METHODS@#Sprague-Dauley (SD) rats were randomly divided into four groups as the following: sham group, hypoxia ischemia brain damage group, curcumin group and solvent control group. The content of MDA in the brain was measured by colorimetry. The expression level of c-fos protein in the cortex tissue was detected by immunohistochemistry. Morphologic and structural changes of neuron cells of the cortex were observed by electron microscopy.@*RESULTS@#The content of MDA was clearly lower in curcumin group than that in the other groups at the same time after HIBD. The expression level of c-fos protein was higher in the curcumin group than that in the other groups (P<0.05). Electron microscopy showed that the morphologic and structural changes of neuron cells of cortex in the curcumin group were reduced.@*CONCLUSION@#Curcumin could significantly decrease the content of MDA, increase the expression level of c-fos protein and reduce the damage of the neuron cells.

ABSTRACT

The antagonism of two strains B1 and B2 of Bacillus spp. against pea root rot pathogen Fusarium oxysporum f. sp. pisi was studied. The result of pairing culture showed that B1 and B2 strains of Bacillus spp. had strong antifungal activity to the pathogen. The colonial color of the pathogen changed from gray to white, aerial hyphas increased and entangled into group after treatment with the cell-free fermentational filtrate of B1 or B2. Observation under optical microscope showed that hyphas and spores of the pathogen swelled and distorted with concentrated cytoplasm after treatment, the spores could not germinate or germinated abnormally . A lot of vesicles appeared at the top of the hyphas, and the hyphas stopped growing and broke finally, their cytoplasm spilled from the cell. The cell-free fermentational filtrate of B1 or B2 strains contained 1795.53?g/mL and 1345.93?g/mL protein respectively, from which two antifungal proteins of 103.5 kD (B1 ) and 127.6 kD (B2) were purified.
